Anum Glasgow, Assistant Professor, Dept. of Biochemistry & Molecular Biophysics, Columbia University, will present a talk titled, "Metabolic tuning of enzyme activity through combinatorial allostery."

ABSTRACT
Allosteric ligand regulation of enzymes that do not undergo dramatic observable structural changes is ubiquitous, but poorly understood. The problem is exacerbated when multiple ligands can combinatorially regulate the enzyme, or when the enzyme is cooperative. Understanding these regulatory mechanisms is a prerequisite for rationally designing therapeutics to target essential enzymes. I will present our efforts to determine the molecular basis for activation and inhibition in an ancient and broadly conserved glycolytic enzyme to determine a unified regulatory mechanism for its ensemble behavior and function, using a combination of X-ray crystallography, hydrogen-deuterium exchange with mass spectrometry, molecular dynamics, Markov state models, and statistical coupling analysis.
